Cytoplasmic Microtubules

چکیده

Small cytoplasmic tubules are present in the interstitial cells and cnidoblasts of hydra. They are referred to here as "microtubules." These tubular elements have an outside diameter of 180 A and an inside diameter of 80 A. By difference, the membranous wall is estimated to be 50 A thick. The maximum length of the microtubules cannot be determined from thin sections but is known to exceed 1.5 micro. In the interstitial cells the microtubules are found in the intercellular bridges, free in the cytoplasm and in association with the centrioles. In the cnidoblast they form a framework around the developing nematocyst and in late stages are related to the cnidocil forming a tight skein in the basal part of the cell. Especially in this cell, confluence of microtubules with small spherical vesicles of the Golgi complex has been observed. It is proposed that these tubules function in the transport of water, ions, or small molecules.

Physarum plasmodia do contain cytoplasmic microtubules!

It has been claimed that the plasmodium of the myxomycete Physarum polycephalum constitutes a very unusual syncytium, devoid of cytoplasmic microtubules. In contrast, we have observed a cytoplasmic microtubule network, by both electron microscopy and immunofluorescence in standard synchronous plasmodia, either in semi-thin sections or in smears, and in thin plasmodia, used as a convenient model...

Microtubules containing acetylated alpha-tubulin in mammalian cells in culture

The subcellular distribution of microtubules containing acetylated alpha-tubulin in mammalian cells in culture was analyzed with 6-11B-1, a monoclonal antibody specific for acetylated alpha-tubulin. Cultures of 3T3, HeLa, and PtK2 cells were grown on coverslips and observed by immunofluorescence microscopy after double-staining by 6-11B-1 and B-5-1-2, a monoclonal antibody specific for all alph...

Role of the cytoplasmic and the intranuclear microtubules on the behavior of pronuclei during conjugation in Paramecium caudatum

Fertilization process of Paramecium caudatum includes a reciprocal exchange of migratory pronuclei across a temporary junction between conjugating cells. To know the role of microtubules on the behavior of pronuclei during conjugation in P. caudatum, the localization of microtubules was investigated by indirect immunofluorescence with a monoclonal anti-a-tubulin antibody. The immunofluorescence...

Flagellar regeneration requires cytoplasmic microtubule depolymerization and kinesin-13.

In ciliated cells, two types of microtubules can be categorized: cytoplasmic and axonemal. It has been shown that axonemal tubulins come from a 'cytoplasmic pool' during cilia regeneration. However, the identity and regulation of this 'pool' is not understood. Previously, we have shown that Chlamydomonas kinesin-13 (CrKin13) is phosphorylated during flagellar regeneration, and required for prop...

Cytoplasmic microtubules and fungal morphogenesis: ultrastructural effects of methyl benzimidazole-2-ylcarbamate determined by freeze- substitution of hyphal tip cells

The effects of methyl benzimidazole-2-ylcarbamate (MBC), one of only a few agents that are active against microtubules of fungi, were analyzed at the ultrastructural level in freeze-substituted hyphal tip cells of Fusarium acuminatum. Nontreated and control cells had numerous microtubules throughout. After just 10 min of exposure to MBC, almost no cytoplasmic microtubules were present, except n...

Cytoplasmic Microtubules in Different Animal Cells

In avian, murine, and human cells fixed with glutaraldehyde, cytoplasmic microtubules 180 to 250 A in diameter and of undetermined length were found. These cytoplasmic microtubules are similar to those described by Ledbetter and Porter in plant cells after the same glutaraldehyde fixation. The cytoplasmic microtubules in animal cells are connected with the satellites of the centrioles and are s...
